Capillary Ion Chromatography: Bisphosphonate Pharmaceuticals

  
  
  
  
  
  

bisphosphonates and excipients in prescription tabletsHere, we discuss a quantitative method for the analysis of bisphosphonates (Wikipedia) and excipients in prescription tablets using newly introduced Capillary Ion chromatography (IC) technique. Bisphosphonates are a group of compounds used as active pharmaceutical ingredients (APIs) for the treatment of bone disorders, Paget’s disease and hypercalcemia.

The key benefit of Capillary IC is that it helps chemists save time, labor, and reduce operating cost, while increasing the productivity and reproducibility of ion analysis. How? Since the Capillary IC system (in this case, Thermo Scientific Dionex ICS-5000 Capillary IC) operates at such low flow rates (~10 µl/min), it is practical to operate a Capillary IC system 24/7, so the system is ready to run samples at any time!  Also, the Eluent Generator module is able to produce eluent automatically from deionized water for up to 18 months under continuous operation, making gradient separations as easy as isocratic.  

In 2008, ScienceDirect.com featured an article, Determination of bisphosphonate active pharmaceutical ingredients in pharmaceuticals and biological material: A review of analytical methods, concluding that for pharmaceutical quality control (QC), IC with conductivity was the choice for analysis because of its simplicity, sensitivity, simultaneous separation of ionic impurities, and the fact that it did not require derivatization.

Here, we present a poster, Quantitative Determination of Bisphosphonate Pharmaceuticals and Excipients by Capillary Ion Chromatography Mass Spectrometry, (downloadable PDF) describing using our Capillary IC system and a capillary anion-exchange column (Thermo Scientific Dionex IonPac™ AS18 column) to achieve chromatographic retention and resolution for target analytes. The derivatization steps are eliminated thus simplifying the workflow and improving method throughput. The detection by suppressed conductivity provides sufficient sensitivity for QC analysis, and the single quadrupole mass spectrometer (Thermo Scientific MSQ Plus™) provides additional selectivity and sensitivity for bisphosphonates in biological fluids.
