HPLC Column for Wide Variety of Pharma Counterions Released!


pharma counterion analysis I am very pleased to feature a newly released mixed-mode HPLC column (Thermo Scientific Acclaim Trinity P2 column) that has been designed to screen a wide range of pharmaceutical counterions, cations, anions, mono- and multi-valent ions in a single analysis!

This particular column, like others in this family of columns, uses our advanced mixed-mode Nanopolymer Silica Hybrid (NSH) technology (downloadable PDF describing the construction of NSH materials). The chemistry is designed to provide spatial separation of the anion exchange and cation regions, resulting in greater resolution and selectivity control. (The image accompanying this post shows the column chemistry.) Also, the hydrophilic surface of the particles makes the this particular column useful as a HILIC column. This means chemists can develop robust, fast, generic methods to improve productivity and reduce cost in pharmaceutical analysis.

The column is featured in Application Note 20868, Simultaneous Determination of Metformin and its Chloride Counterion Using Multi-Mode Liquid Chromatography with Charged Aerosol Detection (downloadable PDF), which describes the determination of metformin (API) and its counterion (chloride) using a simple isocratic method in less than 5 mins. Metformin is an antidiabetic drug used in the treatment of type diabetes. By the way, only metformin has been "conclusively shown to prevent the cardiovascular complications of diabetes." As chloride cannot be detected by UV detection and both metformin and chloride are non-volatile, one of our latest aerosol-based detectors (Thermo Scientific Dionex Corona Veo Charged Aerosol Detector) was used in the method development and is is ideal for this application. Note that this detector can be used with traditional HPLC, UHPLC, and micro LC techniques.

One of our rapid-separation HPLC systems (Thermo Scientific Dionex UltiMate 3000 RSLC system) was used along with our Chromatography Data System (CDS) software (Thermo Scientific Dionex Chromeleon CDS software version 6.8) for instrument control and data management.
